Objective:Seroquel, a SEROQUEL-approved drug, is a unique pharmacologic agent with significant pharmacologic properties. Its pharmacological activity is due to the inhibition of cyclooxygenase-2 (COX-2) and of the synthesis of prostaglandins. COX-2 is a critical regulator of metabolic processes involved in the development of neurodegenerative diseases including neuropathic pain, neuropathic trigeminal neuralgia, and diabetic neuropathy. Therefore, this study aimed to investigate the effect of SEROQUEL on the expression of COX-2 in cultured cells and the changes in expression of COX-2 in cultured cells following exposure to seroquel. The effect of seroquel on the expression of COX-2 in cultured cells was also determined. The expression of COX-2 was measured using real-time PCR.

Methods:The expression of COX-2 was assessed by Western blotting in cultured cells from experimental rats. Seroquel treatment caused significant decreases in the expression of COX-2 compared to that in cells treated for 12 h with a dose of 100 mg/kg body weight. Seroquel treatment also significantly reduced the expression of COX-2 in cultured cells and cultured cells cultured in the presence of seroquel. The expression of COX-2 in cultured cells from experimental rats was decreased by seroquel exposure, and the expression of COX-2 was decreased in cultured cells cultured in seroquel-treated cells. The expression of COX-2 in cultured cells from experimentally treated rats was increased by seroquel exposure. Materials and Methods

Animals

Sixty-two male Wistar rats (12-16 weeks old, female), weighting between 20 and 25 g, were purchased from the National Laboratory Animal Center of the State of New York (North Charles, NY, USA). The animals were housed in a controlled room with 12/12-hour light/light cycles and allowed to acclimate to the experimental diets prior to use.

The rats were anesthetized by an intramuscular injection of chloral hydrate (0.3 mg/kg body weight) and then intraperitoneally (i.p.) intubated.
